Er zijn veel soorten microbiële luchtmonsternemers. De meest gebruikte is de microbiële luchtmonsternemer gebaseerd op de impactmethode op een agarplaat (ook bekend als impactor).
Dit type monsternemer heeft 2 hoofdonderdelen: een luchtpomp en een kopstuk. Het kopstuk bevat een agarplaat. De luchtpomp trekt de lucht door een geperforeerd deksel in het kopstuk. De lucht maakt dan een korte draai boven de agarplaat. Door de centrifugale kracht komen de deeltjes in de lucht in contact met het agaroppervlak en blijven ze plakken. Micro-organismen verplaatsen zich altijd op stof- of aerosoldeeltjes. Ze verplaatsen zich zelden alleen. Door de agarplaat na de bemonstering te incuberen, zullen de micro-organismen zich vermenigvuldigen. Na een bepaalde incubatietijd zijn de clusters van micro-organismen zichtbaar op de plaat en kunnen de zogenaamde kolonievormende eenheden worden geteld. Op basis van het volume van de bemonsterde lucht kun je dan het aantal kolonievormende eenheden berekenen dat per volume lucht aanwezig is. Deze methode wordt het meest gebruikt voor de zogenaamde 'active air sampling' of 'viable particle monitoring'.
Wat zijn de voordelen van een microbiële luchtmonsternemer door impact op agar?
Deze methode heeft een groot voordeel bij routinematige microbiologische kwaliteitscontrole van de lucht. De meest gebruikte parameters hiervoor zijn de totale kiemgetal en het totale aantal gisten en schimmels. Meestal heb je een idee van het aantal kolonievormende eenheden in de lucht en wil je alleen kweekbare micro-organismen meten. Met de microbiologische luchtmonster door impact op agar, heb je een zeer snelle, gemakkelijke en goedkope methode om de luchtkwaliteit te monitoren. Je neemt direct een monster op de plaat, incubeert het en telt het. Kweken is de eenvoudigste en goedkoopste detectiemethode. Het wordt veel gebruikt in verschillende industrieën en richtlijnen.
Wat zegt GMP Annex 1 over luchtmonitoring?
De EU GMP Annex 1 is een richtlijn voor steriele productie (in farmaceutische en biotechnologische productieomgevingen). De EU GMP Annex 1 heeft een sectie die gaat over 'viable en non-viable omgevings- en procesmonitoring'. Luchtbemonstering is een belangrijk onderdeel van dit hoofdstuk. De richtlijn spreekt over 3 soorten luchtbemonstering:
Passieve bemonstering van viable partikels/ bezinkplaten: dit is een simulatie van wat er in werkelijkheid gebeurt. Je opent de agarplaat voor maximaal 4 uur, je sluit hem, incubeert hem en aan het eind kun je de kolonies tellen.
Volumetrische luchtbemonstering / actieve luchtbemonstering van viable particles: dit wordt meestal gedaan met een impactor/microbiële luchtmonsternemer. Je bemonstert maximaal 1000L per plaat met 100l/min of 200l/min in een cleanroomomgeving. Als je continue monitoring toepast volgens de nieuwe GMP Annex 1 met bijvoorbeeld 25l/min, zijn grotere volumes per plaat mogelijk. Daarna incubeer je de plaat en tel je de kolonies. Dit geeft je exacte gegevens over het aantal levensvatbare deeltjes per luchtvolume.
Monitoring van niet-levensvatbare deeltjes: voor dit onderdeel meet je het aantal stofdeeltjes van <0,5 µm en <5µm met behulp van een luchtmonstername-instrument (vaak deeltjestellers genoemd).
Voor alle 3 bovengenoemde soorten luchtmonitoring definieert de EU GMP Annex 1 bepaalde drempelwaarden voor de verschillende cleanroom grades: grade A, B, C en D. Ook de frequentie van de monstername is afhankelijk van de cleanroom grade. Voor zones van klasse A moeten de microbiële luchtmonsters in roestvrij staal worden genomen en worden vaak satellietmonsternemers of sondes gebruikt.
Continuous viable particle monitoring volgens de nieuwe GMP Annex 1
Dit is wat de nieuwe GMP Annex 1 vermeld onder hoofdstuk 9:
9.24 Continuous viable air monitoring in grade A (e.g. air sampling or settle plates) should
be undertaken for the full duration of critical processing, including equipment (aseptic set-up)
assembly and critical processing. A similar approach should be considered for grade B cleanrooms
based on the risk of impact on the aseptic processing. The monitoring should be performed
in such a way that all interventions, transient events and any system deterioration would be
captured and any risk caused by interventions of the monitoring operations is avoided.
De agarplaat zal progressief uitdrogen en zal nutritionele waarde verliezen die nodig is voor sommige micro-organismen. Het gevolg is dat sommige micro-organismen het moeilijk zullen hebben om te vermenigvuldigen waardoor sommige kolonies niet goed zichtbaar zullen worden en mogelijks niet zullen worden meegenomen in de kolonietelling.
Het is nodig om een luchtbemonsteringstoestel te gebrijken met een lager debiet (bijvoorbeeld 25l/min) en om petrischalen te gebruiken met ten minste 30 ml cultuurmedium.
