Wat is een microbiologische luchtmonstername?

Er bestaan verschillende types microbiële luchtbemonsteraars. De meest gebruikte is de impactiebemonsteraar op agarplaten, de zogenaamde impactor.


Dit systeem bestaat uit twee hoofdcomponenten:

  • Een luchtpomp
  • Een bemonsteringskopstuk met een agarplaat


De pomp zuigt lucht via een geperforeerd deksel in het kopstuk. De luchtstroom verandert van richting boven het agaroppervlak en door de centrifugaalkracht slaan de zwevende deeltjes neer op de agar en hechten eraan.


Micro-organismen reizen zelden alleen; ze zijn meestal verbonden met stof- of aerosoldeeltjes. Na de bemonstering wordt de agarplaat geïncubeerd. De micro-organismen vermenigvuldigen zich en vormen zichtbare kolonies, geteld als kolonievormende eenheden (CFU).


Op basis van het bemonsterde luchtvolume wordt het aantal CFU per kubieke meter berekend.


Dit is de meest gebruikte methode voor:

  • Actieve luchtbemonstering
  • Monitoring van levensvatbare deeltjes

Voordelen van impactie op agar

Voor de routinematige microbiële luchtkwaliteitscontrole biedt deze methode grote voordelen.


Veelgebruikte monitoringparameters:

  • Totaal kiemgetal (TVC)
  • Totaal gisten en schimmels
  • In de meeste gecontroleerde omgevingen zijn de verwachte CFU-ranges bekend; het doel is de kweekbare micro-organismen efficiënt te meten.

Het blijft een van de meest praktische en economische microbiologische monitoringmethoden in de farmaceutische, biotech-, voedings- en milieusector.


EU GMP Annex 1 — eisen voor luchtmonitoring

EU GMP Annex 1 regelt de steriele productie in farmaceutische en biotechnologische omgevingen.


Het bevat een specifieke sectie over 'levensvatbare en niet-levensvatbare omgevings- en procesmonitoring'.


Er worden drie types luchtmonitoring gedefinieerd:


1. Passieve bemonstering (settle plates)

  • Agarplaten maximaal 4 uur blootgesteld
  • Simuleert reële contaminatieomstandigheden
  • Platen geïncubeerd en kolonies geteld


2. Volumetrische (actieve) bemonstering

  • Meestal met een impactor
  • Standaard: max. 1000 L per plaat bij 100 L/min
  • Continue monitoring (bijv. 25 L/min) laat grotere volumes toe
  • Levert CFU per luchtvolume


3. Monitoring van niet-levensvatbare deeltjes

  • Meting van deeltjes <0,5 µm en <5 µm
  • Uitgevoerd met deeltjestellers


Grenswaarden en bemonsteringsfrequenties zijn vastgelegd voor klassen A, B, C en D. In Grade A-zones moeten de bemonsteraars in roestvrij staal zijn, vaak met satellietsondes.

Volgens EU GMP Annex 1 §9.24:


Continue monitoring van levensvatbare lucht in Grade A moet gebeuren gedurende de volledige duur van het kritische proces, inclusief de aseptische opstelling en de handelingen.


Voor Grade B geldt een vergelijkbare, risicogebaseerde aanpak.


De monitoring moet:

  • Interventies en tijdelijke gebeurtenissen registreren
  • Achteruitgang van het systeem detecteren
  • Geen extra contaminatierisico introduceren



Bij langdurige luchtbemonstering droogt de agar geleidelijk uit.


Het vochtverlies vermindert de beschikbaarheid van voedingsstoffen, wat kan leiden tot:

  • Remming van de microbiële groei
  • Onderschatting van de CFU
  • Minder betrouwbare data


Hoe uitdroging van de agar voorkomen

Om uitdroging tijdens continue monitoring te beperken:

  • Gebruik een bemonsteraar met een laag debiet (bijv. 25 L/min)
  • Gebruik petrischalen met ≥ 30 mL agar

Deze maatregelen behouden de integriteit van het medium en zorgen voor een betrouwbare kolonietelling.

The TRIO.BAS microbial air samplers have an electrical motor that do not produce particles after the cleaning procedure.
If the same air sampler is transferred in different environments with different levels of microbial contamination (from high to low – e.g. from C to A area), a risk of cross-contamination is possible.
In this case, to avoid this risk of transferring particles from the inside part of the aspirating chamber, it is suggested to adopt those air samplers that include an HEPA filter fixed on the air outlet of instrument.
The purpose of the filter is to stop the potential transfer of particles.
The HEPA filter is positioned externally and therefore its replacement is very simple.

  • Main customers of TRIO.BAS MONO FILTER are pharmaceuticals, cleanrooms and biotech industries. The HEPA filter allows to capture the particulates
  • The data is transferred via Bluetooth between the air sampler and a smartphone or tablet (Android version) and then to a PC or laptop
  • The data may be transferred via cable, too. This is helpful for all companies that, due to internal policy, are not allowed to use wireless transfer
