Qu’est-ce qu’un échantillonneur microbiologique d’air ?

Il existe plusieurs types d'échantillonneurs d'air microbiologiques. Le plus utilisé est l'échantillonneur par impact sur gélose, appelé impacteur.


Ce système comprend deux composants principaux :

  • Une pompe d'aspiration
  • Une tête d'échantillonnage contenant une boîte de gélose


La pompe aspire l'air à travers un couvercle perforé jusqu'à la tête. Le flux change de direction au-dessus de la gélose et, sous l'effet de la force centrifuge, les particules en suspension viennent impacter la gélose et y adhèrent.


Les micro-organismes voyagent rarement seuls : ils sont généralement associés à des particules de poussière ou d'aérosol. Après l'échantillonnage, la gélose est incubée. Les micro-organismes se multiplient et forment des colonies visibles, comptées en unités formant colonies (CFU).


À partir du volume d'air prélevé, on calcule le nombre de CFU par mètre cube.


C'est la méthode la plus courante pour :

  • L'échantillonnage d'air actif
  • La surveillance des particules viables

Avantages de l'impaction sur gélose

Pour le contrôle qualité microbiologique de routine de l'air, cette méthode offre des avantages majeurs.


Paramètres de surveillance courants :

  • Flore aérobie totale (TVC)
  • Levures et moisissures totales
  • Dans la plupart des environnements maîtrisés, les plages de CFU attendues sont connues ; l'objectif est de mesurer efficacement les micro-organismes cultivables.
  • L'impaction sur gélose offre :

​Un échantillonnage direct sur la boîte
​Une incubation simple
​Une détection économique
Une large reconnaissance réglementaire

Elle reste l'une des méthodes de surveillance microbiologique les plus pratiques et économiques dans les secteurs pharmaceutique, biotechnologique, agroalimentaire et environnemental.


EU GMP Annexe 1 — exigences de surveillance de l'air

L'EU GMP Annexe 1 encadre la production stérile en environnement pharmaceutique et biotechnologique.


Elle comporte une section dédiée à la « surveillance environnementale et des procédés, viable et non viable ».


Trois types de surveillance de l'air sont définis :​


1. Échantillonnage passif (plaques de sédimentation)

  • Gélose exposée jusqu'à 4 heures
  • Simule les conditions réelles de contamination
  • Plaques incubées puis colonies comptées


2. Échantillonnage volumétrique (actif)

  • Réalisé généralement avec un impacteur
  • Standard : max. 1000 L par boîte à 100 L/min
  • La surveillance continue (p. ex. 25 L/min) permet des volumes plus élevés
  • Fournit un nombre de CFU par volume d'air


3. Surveillance des particules non viables

  • Mesure des particules <0,5 µm et <5 µm
  • Réalisée avec des compteurs de particules


Des seuils et des fréquences d'échantillonnage sont définis pour les classes A, B, C et D. En zone Grade A, les échantillonneurs doivent être en inox, souvent avec des sondes satellites.​

Selon l'EU GMP Annexe 1 §9.24 :


La surveillance continue de l'air viable en Grade A doit être réalisée pendant toute la durée du procédé critique, y compris la mise en place aseptique et les opérations.


Une approche similaire, fondée sur le risque, s'applique au Grade B.


La surveillance doit :

  • Capter les interventions et les événements transitoires
  • Détecter toute détérioration du système
  • Éviter d'introduire un risque de contamination



Lors d'un échantillonnage d'air prolongé, la gélose se dessèche progressivement.


La perte d'humidité réduit la disponibilité des nutriments, ce qui peut :

  • Inhiber la croissance microbienne
  • Entraîner une sous-estimation des CFU
  • Altérer la fiabilité des données


Comment prévenir le dessèchement de la gélose

Pour limiter le dessèchement lors d'une surveillance continue :

  • Utiliser un échantillonneur à faible débit (p. ex. 25 L/min)
  • Utiliser des boîtes de Pétri contenant ≥ 30 mL de gélose

Ces mesures préservent l'intégrité du milieu et garantissent un comptage de colonies fiable.

The TRIO.BAS microbial air samplers have an electrical motor that do not produce particles after the cleaning procedure.
If the same air sampler is transferred in different environments with different levels of microbial contamination (from high to low – e.g. from C to A area), a risk of cross-contamination is possible.
In this case, to avoid this risk of transferring particles from the inside part of the aspirating chamber, it is suggested to adopt those air samplers that include an HEPA filter fixed on the air outlet of instrument.
The purpose of the filter is to stop the potential transfer of particles.
The HEPA filter is positioned externally and therefore its replacement is very simple.

  • Main customers of TRIO.BAS MONO FILTER are pharmaceuticals, cleanrooms and biotech industries. The HEPA filter allows to capture the particulates
  • The data is transferred via Bluetooth between the air sampler and a smartphone or tablet (Android version) and then to a PC or laptop
  • The data may be transferred via cable, too. This is helpful for all companies that, due to internal policy, are not allowed to use wireless transfer
  • It is possible to work either in manual or automatic mode
  • Body IP 65 certified for protection from dust and water
  • The battery is recharged by a power cable connected directly to the air sampler
  • The use of sterile "Daily Shift" aspirating heads reduces the risk of contamination
