Wat is een PCR-kast?
Een PCR-kast of PCR-werkstation is een verticale laminaire flowkast die is aangepast voor het gebruik van PCR-apparatuur. Het zorgt voor een neerwaartse stroom van gefilterde lucht om je experiment te beschermen tegen kruisbesmetting met luchtverontreinigende stoffen. PCR-kasten hebben altijd een UV-lamp om de kast te ontsmetten van RNA en DNA tussen 2 PCR-cycli.
Een PCR-kast is in principe hetzelfde als een verticale laminaire flowkast, maar met een paar aanpassingen:
- Verminderde hoogte: bij PCR heb je niet veel hoogte nodig. In feite gaat het bij de manipulaties rond PCR vooral om pipetteren, wat op werkbladhoogte gebeurt.
- Geoptimaliseerd voor snelle UV-decontaminatie. UV-ontsmetting is zeer efficiënt om RNA en DNA te vernietigen. Daarom is het handig om een snelle methode te hebben om de UV-lamp te activeren tussen 2 PCR-cycli. Tijdens de UV-ontsmetting moet het schuifraam gesloten zijn om de operator te beschermen. Een PCR-kast heeft een schuifraam met scharnieren dat gemakkelijk gesloten kan worden. Als het schuifraam gesloten is, wordt de UV-lamp automatisch geactiveerd om de UV-ontsmetting te starten.
- Electrical sockets and other items that are not resistant to prolonged exposure to UV radiation are not present in PCR cabinets.
- PCR cabinets also typically have a shelf near the UV lamp. Pipettes can be put on the shelf for decontamination.
Beide kasttypes hebben een UV-lamp en zijn geoptimaliseerd voor een snelle activering van UV-decontaminatie. Een PCR-kast of PCR-werkstation heeft een downflow van gefilterde lucht, die het experiment beschermt tegen kruisbesmetting met de lucht. Een dode luchtkast heeft dit niet. Een dode luchtkast is gewoon een kast met een UV-lamp en biedt geen enkele vorm van bescherming van uw experiment tegen luchtverontreinigingen wanneer de kast open is.
