What are the applications of upright microscopes?
In life sciences and cell biology, upright microscopes are used for phase contrast, brightfield, darkfield, differential interference contrast (DIC), polarization and fluorescence microscopy of slide samples.
They are also suited to the microscopy of fixed cells and tissue samples.
How does an upright microscope work?
In an upright microscope, the light source and condenser sit below the stage — and therefore below the specimen. Light passes through the sample from underneath, and you observe it from above through the ocular lens.
With an inverted microscope, gravity makes cells sink and adhere to the coverslip. With an upright microscope used for cell biology, the sample is instead pressed between the coverslip and the slide. This lets you view and analyze it clearly from above.
